Yes. Any number of such setups are possible. The simples having three
Squids, one main Squid where the clients connect and all caching are
done, and two virtual upstream peers (on the same box) bound to each of
the ISP interfaces. The main squid is then configured to use the other
two in round-robin mode.

Or if you have some selection criteria based on the clients, you could
use only two Squids, and cache_peer_access to route the requests which
should go by the "alternate" path to the other non-caching Squid.
